TransCode Therapeutics, a clinical-stage biotechnology firm based in Boston, recently concluded its Phase 1a dose-escalation clinical trial for TTX-MC138, an RNA therapeutic aimed at combating cancer. The study successfully achieved its primary safety endpoints, demonstrating the drug's safety and effectiveness in stabilizing advanced metastatic cancer in patients, indicating a promising path forward for its continued development.

The Phase 1a trial specifically focused on evaluating the safety, tolerability, and pharmacokinetic profile of TTX-MC138 within a patient group that had undergone extensive prior treatments. Across all dosage cohorts, the therapeutic was well-received, with no dose-limiting toxicities observed, suggesting a favorable safety profile. Out of 14 evaluable patients, a notable 64% exhibited stable disease for a minimum of six months. One patient with metastatic thyroid cancer showed a significant reduction in thyroglobulin levels, a key cancer marker, maintaining disease stability for 12 months and continuing participation in the study. These findings highlight the drug's potential for durable disease control in individuals with highly advanced forms of cancer.

The successful completion of the dose-escalation phase allowed TransCode to pinpoint 4.8 mg/kg as the recommended dosage for its upcoming Phase 2a study. This next phase will concentrate on assessing TTX-MC138's efficacy in patients with circulating tumor DNA (ctDNA) positive colorectal cancer, particularly after they have received curative-intent therapy. TTX-MC138 is designed to inhibit microRNA-10b (miR-10b), a critical biomarker and driver in numerous metastatic cancers, representing a novel approach to cancer treatment.
